How is Endoplasmic Reticulum Stress Recognised

A homeostatic pathway known as the Unfolded Protein Response (UPR) adapts the protein folding and processing machinery of the early secretory pathway to the burden of unfolded protein in the compartment (so-called ER stress). A good deal is known about the effectors of the UPR, but the upstream signal(s) that triggers their activation is less well understood. In this talk I will review the prevailing (and sometimes conflicting) ideas on this topic. Focussing on IRE1, the ancient and most conserved UPR effector I will present some recent insights into the structural basis of this upstream process, gleaned from the study of a vertebrate-specific isoform of IRE1, expressed selectively in mucin-producing cells (BioRxiv 10.1101/2025.04.14.648677).
