Mechanics of cell integration in vivo
During embryonic development, regeneration and homeostasis, precursor cells have to integrate into their target tissues where they ultimately execute their function. Despite a significant body of research on cells' interplay with their passive surroundings, little is known about how cells perceive and interpret active tissue environments. Here, using quantitative in vivo imaging and molecular perturbations, together with a theoretical model, we reveal that intercalating multiciliated cell (MCC) progenitors in the Xenopus embryo form dynamic filopodia that probe and pull at the vertices of the overlying epithelial sheet to identify a susceptible cellular environment for their integration into the tissue. Moreover, we report a novel mechanotransducive function for lipolysis-stimulated lipoprotein receptor (LSR) protein in this process. Remarkably, we find that pulling forces equip the MCCs to remodel junctions in the overlying tissue, enabling them to generate a favorable environment for their own integration. Altogether, our work reveals how cells dynamically perceive their active environment in vivo, and emphasizes a cell’s ability to both read mechanical cues from and modify its surroundings.
